Hi Holger,
Sometimes this can merely be a display problem. To check this out, you can enlarge the text on the screen and see if this appearance persists. Ultimately, printing out a page to see if this appearance is there on the printed page would be the final test.
If it is there even when printed you have to go back to Scribus and check out these letters to find out what happened. Using paragraph styles can make this easier than fiddling with individual letters.
I don't think the size of the file makes any difference.
